A new innovated and cheep model in building artificial reefs OceanDocs
Mahmoud, M.A.; Ammar, M.S.A..
A novel and cheep approach for building artificial reefs was designed in such a way that sediments in an area of high sedimentation rate will be utilized in building the artificial substrate with no further sediment accumulation afterwards. The model consists of several steel cylinders which are soldered together in such a way to form a reef flat, a gentle slope and a steep slope, with two layers of narrow opening meshes (a lower plastic and an upper metallic one). Within a short periode (3 months), a substrate layer of a total of ~ 1 cm was formed between the two meshes, this layer increased to ~2 cm after 9 months. Transplants of the three species Stylophora pistillata, Echinopora gemmacea and Montipora spongiosa grow better when the attaching surface is...

Adaptation of copper community tolerance levels after biofilm transplantation in an urban river ArchiMer
Fechner, Lise C.; Versace, Francois; Gourlay-france, Catherine; Tusseau-vuillemin, Marie-helene.
The Water Framework Directive requires the development of biological tools which can act as early-warning indicators of a sudden increase (accidental pollution) or decrease (recovery due to prevention) of the chemical status of aquatic systems. River biofilms, which respond quickly to modifications of environmental parameters and also play a key part in the functioning of aquatic ecosystems, are therefore good candidates to monitor an increase or a decrease of water pollution. In the present study, we investigated the biological response of biofilms transplanted either upstream (recovery) or downstream (deterioration of exposure levels) the urban area of Paris (France). Both modifications of Cu community tolerance levels and of global bacterial and...

Beneficial Effect of Hyperbaric Oxygen Therapy on the Follicular Survival in the Mouse Ovary after Transplantation OAK
HAYASHI, Satomi; ASANO, Tomoyoshi; KAKIZAKI, Ryujiro; SUZUKI, Hiroshi; 鈴木, 宏志.
A large proportion of follicles are lost during the initial ischemia that occurs after transplantation of ovarian tissues. Thus, the effect of hyperbaric oxygen therapy (HBO) on the follicular loss of ovarian tissues after transplantation was examined in mice. Ovarian slices from ICR mice were transplanted under the kidney capsule in ovariectomized ICR. Hyperbaric oxygen with 100% oxygen was initiated for 30 min at 2.5 atmospheres absolute immediately after transplantation, and this treatment was repeated at 48-h intervals for 2 weeks. The number of follicles was dramatically reduced at 2 weeks post transplantation. However, HBO was significantly effective in enhancing the survival of transplanted ovarian follicles. The survival rates of primordial and...

Biology and clinical utilization of mesenchymal progenitor cells BJMBR
Minguell,J.J.; Conget,P.; Erices,A..
Within the complex cellular arrangement found in the bone marrow stroma there exists a subset of nonhematopoietic cells referred to as mesenchymal progenitor cells (MPC). These cells can be expanded ex vivo and induced, either in vitro or in vivo, to terminally differentiate into at least seven types of cells: osteocytes, chondrocytes, adipocytes, tenocytes, myotubes, astrocytes and hematopoietic-supporting stroma. This broad multipotentiality, the feasibility to obtain MPC from bone marrow, cord and peripheral blood and their transplantability support the impact that the use of MPC will have in clinical settings. However, a number of fundamental questions about the cellular and molecular biology of MPC still need to be resolved before these cells can be...

The cryopreservation of ovarian tissues is a technology with significant potential for the preservation of the genetic resource materials of working dogs, including guide dogs for the blind. However, no attempt has been reported on cryopreservation of the canine ovary. Thus, we evaluated a vitrification method for cryopreservation of canine ovaries and determined the potential functionality of vitrified-warmed canine ovaries by means of transplantation into non-obese diabetic-severe combined immunodeficiency (NOD-SCID) mice. All ovarian tissues cryopreserved by vitrification were morphologically normal in terms of histology. Cryopreserved ovaries were transplanted into the ovarian bursa of the NOD-SCID mice, and the xenografts were recovered from 23 of 23...

Detection of cytomegalovirus infections by PCR in renal transplant patients BJMBR
Costa,S.C.B.; Miranda,S.R.P.; Alves,G.; Rossi,C.L.; Figueiredo,L.T.M.; Costa,F.F..
Cytomegalovirus (CMV) is the single most important infectious agent affecting recipients of organ transplants. To evaluate the incidence and the clinical importance of CMV infection in renal transplants in Brazil, 37 patients submitted to renal allograft transplants were tested periodically for the presence of cytomegalovirus DNA in urine using the polymerase chain reaction (PCR), and for the presence of IgM and IgG antibodies against CMV by enzyme-linked immunosorbent assay (ELISA) and indirect immunofluorescence (IIF). The PCR-amplified products were detected by gel electrophoresis and confirmed by dot-blot hybridization with oligonucleotide probes. Thirty-two of the 37 patients (86.4%) were positive by at least one of the three methods. In six patients,...

Development of two potential diagnostic monoclonal antibodies against human cytomegalovirus glycoprotein B BJMBR
Jun,Fan; Minhuan,Li; Yadan,Ma; Yaping,Huang; Hanying,Liang; Jianhua,Hu; Hangping,Yao; Weihang,Ma.
Human cytomegalovirus glycoprotein B (gB) represents a target for diagnosis and treatment in view of the role it plays in virus entry and spread. Nevertheless, to our knowledge, rare detection of a gB antigen has been reported in transplant patients and limited information is available about diagnostic gB monoclonal antibodies (mAbs). Our aim was to develop gB mAbs with diagnostic potential. Hydrophilic gB peptides (ST: amino acids 27-40, SH: amino acids 81-94) of favorable immunogenicity were synthesized and used to immunize BALB/c mice. Two mAbs, named ZJU-FH6 and ZJU-FE6, were generated by the hybridoma technique and limited serial dilution and then characterized by indirect ELISA, Western blotting, immunoprecipitation, and immunohistochemical staining....

Dynamics of Forage Production in Pasture-woodlands of the Swiss Jura Mountains under Projected Climate Change Scenarios Ecology and Society
Silvopastoral systems of the Swiss Jura Mountains serve as a traditional source of forage and timber in the subalpine vegetation belt, but their vulnerability to land use and climate change puts their future sustainability at stake. We coupled experimental and modeling approaches to assess the impact of climate change on the pasture-woodland landscape. We drew conclusions on the resistance potential of wooded pastures with different management intensities by sampling along a canopy cover gradient. This gradient spanned from unwooded pastures associated with intensive farming to densely wooded pastures associated with extensive farming. Transplanted mesocosms of these ecosystems placed at warmer and drier conditions provided experimental evidence that...

Ectopic development of skeletal muscle induced by subcutaneous transplant of rat satellite cells BJMBR
Fukushima,M.G.; Furlan,I.; Chiavegatti,T.; Kiyomoto,B.H.; Godinho,R.O..
The present study analyzes the ectopic development of the rat skeletal muscle originated from transplanted satellite cells. Satellite cells (10(6) cells) obtained from hindlimb muscles of newborn female 2BAW Wistar rats were injected subcutaneously into the dorsal area of adult male rats. After 3, 7, and 14 days, the transplanted tissues (N = 4-5) were processed for histochemical analysis of peripheral nerves, inactive X-chromosome and acetylcholinesterase. Nicotinic acetylcholine receptors (nAChRs) were also labeled with tetramethylrhodamine-labeled alpha-bungarotoxin. The development of ectopic muscles was successful in 86% of the implantation sites. By day 3, the transplanted cells were organized as multinucleated fibers containing multiple clusters of...

Electrophysiological properties of rat nodose ganglion neurons co-transplanted with carotid bodies into the chick chorioallantoic membrane Biol. Res.
Eugenín,Jaime; Eyzaguirre,Carlos.
The electrophysiological properties of nodose ganglion neurons were evaluated immediately after removing nodose ganglia from young adult rats and 3 to 10 days after nodose ganglia implantation _either alone or co-implanted with carotid bodies_ onto the chick chorioallantoic membrane. Implanted and co-implanted nodose neurons were less excitable than acutely recorded nodose neurons. Co-implanted neurons also showed reduced amplitudes for both action potentials and spike after-hyperpolarizations relative to those found in acutely recorded nodose ganglion neurons and a smaller time constant (τ) than that found in implanted neurons. In addition, no spontaneous activity was recorded from nodose ganglion neurons co-implanted with carotid bodies during...

Etude d'une prolifération hémocytaire chez la moule Mytilus edulis ArchiMer
Noel, Danièle.
Hemic proliferative disease in the bay mussel, Mytilus edulis, has been studied following mortalities in Washington State (USA). The disease has been reproduced in the labortory. The establishment of this experimental model led to study tumoral characteristics of the atypic hemocytes : ultrastrure, in vitro cultivation, transplantation. The infectious etiology of this tumor has been verified. It has been hypothesized to result from a viral infection since retrovirus-like particules have been observed following induction by bromodeoxyuridine. Specific monoclonal antibodies against atypic hemocytes have been obtained and characcterized

The effect of cryopreservation and subsequent xenotransplantation on the follicular reserve of the canine ovary by using non obese diabetic-severe combined immunodeficient (NOD-SCID) mice was examined. Vitrified-warmed canine ovarian tissues were placed into the ovarian bursa of mice, and then were removed and subjected to histological examination at 4 weeks after the transplantation. Over 30% of primordial follicles and 65% of early primary follicles survived after cryopreservation. However, regardless of breed or age, percentages of survived primordial follicles and early primary follicles after the transplantation ranged from 0-7% and from 0-15%, respectively. These results indicate that the majority of primordial follicles and early primary follicles...

Health care-related infections in solid organ transplants BJID
Sola,A.F.; Bittencourt,A.R.C.; Guerra,C.M.; Godoy,H.L.; Medeiros,E.A.S..
The health care-related infections are well-known in a critical care setting, but reports of those infections in solid organ transplanted patients are scarce. We developed a study of retrospective cohort in a tertiary teaching hospital for 14 months. Eighty-one patients underwent solid organ transplants. The global incidence of health care-related infection was 42.0%. Fifteen percent of the cases were occurrences of surgical site infections, 14.0% pneumonias, 9.0% primary blood stream infections, 4.0% urinary tract infections and 2.0% skin infection. The most prevalent etiologic agents were K. pneumoniae (8.6%), P. aeruginosa (7.4%); A. baumannii (5.0%) and S. aureus (2.5%). Mortality was 18.0%, none of then related to health care infections. The high rate...

Hematopoietic stem cell transplantation: clinical use and perspectives Biol. Res.
Barriga,Francisco; Ramírez,Pablo; Wietstruck,Angélica; Rojas,Nicolás.
Hematopoietic stem cell transplantation is the accepted therapy of choice for a variety of malignant and non-malignant diseases in children and adults. Initially developed as rescue therapy for a patient with cancer after high doses of chemotherapy and radiation as well as the correction of severe deficiencies in the hematopoietic system, it has evolved into an adoptive immune therapy for malignancies and autoimmune disorders. The procedure has helped to obtain key information about the bone marrow environment, the biology of hematopoietic stem cells and histocompatibility. The development of this new discipline has allowed numerous groups working around the world to cure patients of diseases previously considered lethal. Together with the ever growing...

Heterotopic transplantation of glycerin-preserved trachea: effect of respiratory epithelium desquamation on acute rejection BJMBR
Saueressig,M.G.; Edelweiss,M.I.A.; Souza,F.H.; Moreschi,A.H.; Savegnago,F.L.; Macedo Neto,A.V..
An effective preservation method and decreased rejection are essential for tracheal transplantation in the reconstruction of large airway defects. Our objective in the present study was to evaluate the antigenic properties of glycerin-preserved tracheal segments. Sixty-one tracheal segments (2.4 to 3.1 cm) were divided into three groups: autograft (N = 21), fresh allograft (N = 18) and glycerin-preserved allograft (N = 22). Two segments from different groups were implanted into the greater omentum of dogs (N = 31). After 28 days, the segments were harvested and analyzed for mononuclear infiltration score and for the presence of respiratory epithelium. The fresh allograft group presented the highest score for mononuclear infiltration (1.78 ± 0.43, P <=...

Implantation of muscle satellite cells overexpressing myogenin improves denervated muscle atrophy in rats BJMBR
Shen,H.; Lv,Y.; Shen,X.Q.; Xu,J.H.; Lu,H.; Fu,L.C.; Duan,T..
This study evaluated the effect of muscle satellite cells (MSCs) overexpressing myogenin (MyoG) on denervated muscle atrophy. Rat MSCs were isolated and transfected with the MyoG-EGFP plasmid vector GV143. MyoG-transfected MSCs (MTMs) were transplanted into rat gastrocnemius muscles at 1 week after surgical denervation. Controls included injections of untransfected MSCs or the vehicle only. Muscles were harvested and analyzed at 2, 4, and 24 weeks post-transplantation. Immunofluorescence confirmed MyoG overexpression in MTMs. The muscle wet weight ratio was significantly reduced at 2 weeks after MTM injection (67.17±6.79) compared with muscles injected with MSCs (58.83±5.31) or the vehicle (53.00±7.67; t=2.37, P=0.04 and t=3.39, P=0.007, respectively). In situ kinetics of mercury bioaccumulation in mytilus galloprovincialis estimated by transplantation experiments ArchiMer
Casas, Stellio; Cossa, Daniel; Gonzalez, Jean-louis; Bacher, Cedric; Andral, Bruno.
Monitoring coastal contamination by trace metals pollution using mollusks bivalves as quantitative bioindicators is widely performed in many international biomonitoring programs. For this purpose, studying mercury dynamic in marine mussels is a reliable tool. In situ experiments on uptake and elimination kinetics were conducted in three Mediterranean sites, chosen on the basis of their contamination levels. Mussels were transplanted from a clean area to a highly contaminated one and then transferred back to a clean site. This approach makes it possible to define bioaccumulation factors and uptake and elimination rates consistently with the real environmental conditions.

Lymphocyte subpopulations during cytomegalovirus disease in renal transplant recipients BJMBR
Castro,S.M.; Sporleder,H.; Schröeder,R.; Santos,A.; Garcia,V.; Neumann,J.; Costa,S.C.B..
We have determined the number of circulating T, B and natural killer cells in renal transplant recipients in order to detect changes during cytomegalovirus (CMV) infections. Serial blood samples were taken from 61 patients on standard triple immunosuppression therapy (cyclosporin A, azathioprine and prednisone). Using two-color flow cytometry analysis, the absolute number of CD3+, CD4+, CD8+, CD19+, CD3+HLA-DR+ and CD16+56+ cells was determined. Forty-eight patients (78.7%) developed active CMV infection, and all of them subsequently recovered. Twenty of the infected patients (32.8%) presented symptoms compatible with CMV disease during the infectious process. The number of lymphocytes and their main subpopulations were normal before the onset of CMV...
